Punicalagin, an active pomegranate component, is a new inhibitor of pdia3 reductase activity

Background: Polyphenolic compounds isolated from pomegranate fruit possess several pharmacological
activities including anti-inflammatory, hepatoprotective, antigenotoxic and anticoagulant activities. The
present work focuses the attention on PDIA3 interaction with punicalagin and ellagic acid, the most
predominant components of pomegranate extracts. PDIA3, a member of the protein disulfide isomerase
family involved in several cellular functions, is associated with different human diseases and it has the
potential to be a pharmacological target.

A comparative analysis of secreted protein disulfide isomerases from the tropical co-endemic parasites Schistosoma mansoni and Leishmania major

The human parasites Schistosoma mansoni and Leishmania major are co-endemic and a major threat to human health. Though displaying different tissue tropisms, they excrete/secrete similar subsets of intracellular proteins that, interacting with the host extracellular matrix (ECM), help the parasites invading the host. We selected one of the most abundant proteins found in the secretomes of both parasites, protein disulfide isomerase (PDI), and performed a comparative screening with surface plasmon resonance imaging (SPRi), looking for ECM binding partners.

Pdia3 expression in glioblastoma modulates macrophage/microglia pro-tumor activation

The glioblastoma (GB) microenvironment includes cells of the innate immune system identified as glioma-associated microglia/macrophages (GAMs) that are still poorly characterized. A potential role on the mechanisms regulating GAM activity might be played by the endoplasmic reticulum protein ERp57/PDIA3 (protein disulfide-isomerase A3), the modulation of which has been reported in a variety of cancers. Moreover, by using The Cancer Genome Atlas database, we found that overexpression of PDIA3 correlated with about 55% reduction of overall survival of glioma patients.

Influence of ellagitannins extracted by pomegranate fruit on disulfide isomerase PDIA3 activity

Pomegranate fruit is a functional food of high interest for human health due to its wide range of phytochemicals with antioxidant properties are implicated in the prevention of inflammation and cancer. Ellagitannins, such as punicalagin and ellagic acid, play a role as anti-atherogenic and neuroprotective molecules in the complex fighting against the degenerative diseases.
