We have developed a three-dimensional skeletal muscle construct, the eX-vivo Muscle Engineered Tissue (X-MET), derived from murine skeletal muscle primary cultures containing satellite cells, fibroblasts, and endothelial cells. X-MET closely mimics native skeletal muscle architecture, providing an ideal in vitro model to study muscle homeostasis, differentiation, and muscle–nerve interactions in physiological and pathological conditions. Building on this platform, our current research integrates stem cell biology and biomaterial engineering to create 3D skeletal muscle models with neuronal networks. These -specific 3D culture system replicates the neuromuscular system, offering valuable tools to investigate neuromuscular development, maintenance, and diseases.
